手性金属卟啉催化氧化制备埃索美拉唑钠工业化工艺

摘    要:基于埃索美拉唑钠是一种性能优异的质子泵抑制剂,是治疗消化性溃疡的有效药物,采用手性金属卟啉催化剂催化氧化的新工艺,实现埃索美拉唑钠的工业化生产,目标产物经过核磁共振谱,质谱和比旋光度证实,并研究反应时间、溶剂、温度及催化剂用量对工艺的影响。研究结果表明:由2-氯甲基-3,5-二甲基-4-甲氧基吡啶盐酸盐和5-甲氧基-2-巯基苯并咪唑在质量分数为20%的NaOH水溶液和TBAB作用下生成奥美拉唑硫醚,再由奥美拉唑硫醚在氧气和NaOH作用下高效合成埃索美拉唑钠;当奥美拉唑硫醚合成埃索美拉唑钠的反应时间为3.0 h,溶剂为甲苯,反应温度为80 ℃,奥美拉唑硫醚与手性金属卟啉摩尔比为1:(3.7×10-5)时,反应产率和对映选择性最高,大规模工业化生产的总收率可达60%,对映体过量百分比高达98.9%。

Abstract: Sodium esomeprazole is a kind of excellent proton pump inhibitor, used as an effective drug for peptic ulcer disease. A new industrialized synthetic technology of sodium esomeprazole catalyzed by chiral metalloporphyrin was reported. The target product was approved by 1H NMR, MS and polarimetry. The effect of reaction time, reaction temperature, and molar ratio of catalyst and solvent were investigated. The results show that sodium esomeprazole can be obtained with high yield from oxidation by oxygen in the NaOH solution and ufiprazole, synthesized by the addition reaction between2-chloro-3,5-dimethyl-4-methoxypyridine hydrochloride and 5-methoxy-2-mercaptobenzimidazole in the combination of 20% NaOH and TBAB. Synthesis of esomeprazole sodium used by ufiprazole hasthe highest yield and enantioselectivity when reaction time is 3.0 h, the solvent is toluene, the reaction temperature is 80 ℃ and the ratio of omeprazole sulfide and chiral metal porphyrin is 1:(3.7×10-5).The total industrial yield is 60% and the enantioselectivity is 98.9%.
