利用Minitab优化耐高温淀粉酶发酵培养条件

摘    要:利用Minitab软件中的Plackett-Burman实验设计和响应面分析法,对耐高温α-淀粉酶产生菌Bacillus subtilis C1的发酵培养条件进行优化。在单次单因子法得出的适宜条件基础上,综合考虑所有影响因素,对培养条件进行统计分析,得到Bacillus subtilis C1产耐高温α-淀粉酶的最佳培养条件。研究结果表明:优化培养基的组成(200 mL)为麸皮2.16 g,棉粕粉1.98 g,酵母粉0.40 g,NaCl 1.00 g,CaCl2 0.40 g,淀粉0.10 g。培养基优化后的酶活为2 329 U/mL,约是培养基优化前酶活的12.55倍,酶活得到显著提高。

Abstract: The optimum medium for Bacillus subtilis C1 was systematically studied with Minitab software. On the basis of single factor experiments, the prime factors affecting enzyme activity were selected by means of Plackett-Burman design. And then, the factors were optimized by response surface analysis. The results show that the optimal medium composition in 200 mL culture medium is as follows: wheat bran 2.16 g, cotton seed meal 1.98 g, yeast extract 0.40 g, NaCl 1.00 g, CaCl2 0.40 g, starch 0.10 g. Enzyme activity under the optimal culture conditions is 2 329 U/mL, which is 12.55 times of that of the original medium.
