细柱五加叶中抗HMGB1总三萜活性物质的闪式提取工艺

摘    要:采用闪式提取,通过二次通用旋转实验设计优化细柱五加叶中三萜类物质的提取工艺,以综合评分为指标,考察乙醇浓度、提取时间、溶剂用量3个主要因素对工艺的影响;确定出最佳提取工艺,用UV法检测提取物中的总三萜成分含量。实验结果表明最佳提取工艺为:53%(体积分数)乙醇为溶剂,闪式提取175 s;该工艺稳定,重现性好(相对标准偏差为0.77%),操作简单、省时,以五加皂苷为对照品,饱和NaHCO3溶液为溶剂,以218 nm为最适波长时,线性范围为0.01~0.07 g/L(相关系数为0.999 6),加样回收率为98.2%(相对标准偏差为1.23%)。

Abstract:

Smashing tissue extraction was used to distill the active triterpenoids of anti-HMGB1 from AGS, and the quadratic general rotary unitized design was used to optimize the extraction process. Taking the overall score as the target, the influences of ethyl alcohol concentration, the extraction time and the solvent amount on extraction process were studied. Finally, the optimal extraction technological conditions were determined. The triterpenoids in extraction was determined by UV. The results show that the optimal condition is taking 53% ethyl alcohol as the solvent and distilling for 175 s. This craft is stable and the reproduction quality is good (the relative standard deviation (RSD) is 0.77%), also the operation is simple and takes shorter time. Using acankoreanogenin A as comparison, saturation NaHCO3 as solvent and the most suitable wavelength is 218 nm, the liner range is 0.01-0.07 g/L (R=0.999 6) and the recovery of acankoreanogenin A is 98.2% (RSD is 1.23%).
