摘 要:通过化学方法合成了嵌合型反义寡核苷酸(ODN-EDTA·M).这种反义寡核苷酸在基因调控及表达、基因治疗中有重要作用,它可以在特定部位切割双链DNA,也可以与双链DNA形成稳定的三链体,抑制目的基因的表达.
Abstract: The synthesis and modification of antisense oligodeoxyribonucleotides (ODN)is described.EDTA has been attached covalently to the 5' terminus of the ODN sequence via an ethylenediamine linker,then chelated with the metal ions such as Fe (Ⅱ),Co(Ⅱ)or Cu(Ⅱ).HPLC is used to separate and purify this kind of antisense modified ODN. By using this routine chemical modification method, EDTA can be linked to the 5' end of ODN of any desired length and sequence.
