西南印度洋深海重金属抗性菌的分离鉴定与多样性分析

摘    要:利用大洋第21航次采集的西南印度洋中脊热液区的环境样品进行了重金属抗性菌的富集分析鉴定与多样性分析。从5个样品中通过Cu,Zn,Cd和Cr 4种重金属富集,平板分离、REP-PCR和16S rDNA鉴定,最终分离得到51株细菌,属于24个属39个种,多样性非常丰富。其中,变形菌纲γ亚纲占绝对优势(48%),其余4类占较小的比例,其中,变形菌纲门α亚纲占14%,放线菌门Actinobacteria占18%,厚壁菌门Firmicutes占12%,拟杆菌门Bacteroidetes占8%。在γ亚纲中Halomonas属和Idiomarina属占大多数,且在大部分站点中都有分布。此外,Nitratireductor,Salegentibacter,Pseudidiomarina,Belliella,Marinimicrobium,Cellulosimicrobium和Joostella是作为重金属抗性菌的首次报道。

Abstract: The bacteria of heavy metal tolerance were isolated and subjected to diversity analysis from the hydrothermal field samples of Southwest Indian Ridge (SWIR) in 21st scientific voyage. Fifty-one strains of heavy metal resistance screened with Cu, Zn, Cd and Cr were obtained from 5 samples of SWIR, which belonged to 39 species of 24 genera identified based on REP-PCR and 16S rDNA, representing a high diversity. Among them, γ-proteobacteria occupies the majority (48%), other 4 subclasses exhibit as minorities including α-proteobacteria (14%), Actinobacteria (18%), Firmicutes (12%) and Bacteroidetes (8%). Bacteria of genus Halomonas and Idiomarina are most frequently isolated. Moreover, bacteria of the following genera including Nitratireductor, Salegentibacter, Pseudidiomarina, Belliella, Marinimicrobium, Cellulosimicrobium and Joostella are the first report as heavy metal resistant bacteria.
