嗜酸氧化亚铁硫杆菌的保藏方法

摘 要:对几种适用于中短期保藏嗜酸氧化亚铁硫杆菌的保藏方法进行对比研究。研究结果表明:在多种保藏方法中,采用单蒸水悬浮细菌(pH=6.8),在4℃保藏效果最佳,与传统的传代保藏效果接近;以稀硫酸(pH=2.0),悬浮在4℃、浓缩静置和浓缩菌液用石蜡油密封在4℃保藏的效果次之;而用9 K基本盐溶液(pH=2.0),悬浮在4℃的保藏效果最差;采用不同保藏方法处理后,该菌的生长迟滞期都有不同程度的延长;而营养缺乏、pH值为7左右、无氧、低盐浓度的环境有利于菌种的保藏。
Abstract: Several methods of short-term preservation ofAcidithiobacillus ferrooxidanswere investigated. The result shows that among these preservation methods, the distilled water suspension (pH=6.8, 4℃)gives the best effect, the sulfuric acid suspension (pH=2.0, 4℃) and concentrate produces an inferior effect at 4℃, and the 9K basic salt aqua suspension (pH=2.0,4℃) shows the worst effect. Furthermore, all the preservation methods extend the lag period of the germ more or less. Nutrition deficiency, pH≈7, anoxic condition and low salinity are propitious to preserve A cidithiobacillus ferrooxidans.
